Mercaptans, or thiols, are organic compounds similar to alcohols in which the oxygen atom has been replaced by a sulfur atom; indeed, their general formula is R-SH. A common characteristic of almost all mercaptans is their intense unpleasant odor. Simple mercaptans are added to methane as "odorizers" to detect possible leaks. They are synthesized by treating alkyl halides with excess hydrosulfide ions HS-: R-X + HS- → R-SH + X-.
The oxidation of a thiol is a reversible process that leads to the formation of disulfides R-S-S-R: the disulfide bond is very important in some proteins; for example, hair can be straight or wavy as a consequence of structural variations due to disulfide bridges.
Like alcohols, their anions R-S- undergo nucleophilic addition and nucleophilic substitution reactions, but thiolates are better nucleophiles compared to alkoxides, since the larger size of sulfur compared to oxygen allows for the delocalization of the negative electric charge over a greater surface area and also fits the attack site better.
Due to the better electronic distribution of the thiolate ion, thiols are more acidic than their corresponding alcohols. This explains the pKa value of around 8-9 compared to that of alcohols (pKa around 17).
For the same reason, they are more reactive in nucleophilic reactions. A significant example is the use of 1,2-ethanedithiol for the protection of the carbonyl group of aldehydes and ketones in organic syntheses.
